microRNA-138 suppressed proliferation and invasion of gastric cancer via RhoC

چکیده
Gastric cancer (GC) is the fourth most predominant malignancy worldwide and remains the second most common cause of cancer-related death globally. Recent studies indicated that miRNAs have been involved in the tumorigenesis pathways in GC. In this study, we investigated the effect and molecular mechanism of miR-138 in human GC. The effects of miR-138 on the GC cell growth and invasion were first studied. Then the correlation of miR138 with RhoC was confirmed by dual luciferase reporter assay. We found that the overexpression of miR-138 resulted in inhibition of GC growth and reversion of invasion. RhoC was identified as the target of miR-138. Altogether, we conclude that miR-138 can modulate gastric cancer cell growth and invasion by suppressing the expression of RhoC and the activation of PI3K/Akt pathway. These data represent a crucial step towards the understanding of the novel roles and molecular mechanism of miR-138, RhoC in GC progression. Therefore, miR-138 may serve as a useful therapeutic agent for miRNA-based GC therapy.

Objective(s):MiRNAs have been proposed to be key regulators of tumorigenesis, progression and metastasis. However, their effect and prognostic value in gastric cancer is still poorly known. Materials and Methods: Gastric cancer cell lines were cultured. Tissue samples obtained from 36 gastric cancer patients were used for quantitative real-time PCR (qRT-PCR) analysis. The tissue microarrays (T...
متن کاملREG3A promotes the proliferation, migration, and invasion of gastric cancer cells
The mechanism underlying the metastasis of gastric cancer (GC) cells remains elusive. REG3A is considered an oncogene in various cancers, but in GC its role is unclear. Here, we report that the expression of REG3A was significantly increased in the tumor tissues of patients with GC compared with the matched normal tissues. Knockdown of REG3A induced by specific small interfering RNA (siRNA) sig...

Epithelial ovarian cancer (EOC) is the most lethal of the gynecological malignancies. Exploring the molecular mechanisms and major factors of invasion and metastasis could have great significance for the treatment and prognosis of EOC. Studies have demonstrated that microRNA 106b (miR-106b) may be a promising therapeutic target for inhibiting breast cancer bone metastasis, but the role of miR-1...

متن کاملmicroRNA-9 Suppresses the Proliferation, Invasion and Metastasis of Gastric Cancer Cells through Targeting Cyclin D1 and Ets1
Recent evidence shows that altered microRNA-9 (miR-9) expression is implicated in the progression of gastric cancer. However, the exact roles and underlying mechanisms of miR-9 in the proliferation, invasion and metastasis of gastric cancer still remain unknown. In this study, miR-9 was found to be down-regulated and inversely correlated with the expression of cyclin D1 and v-ets erythroblastos...
